Function of pyridoxal phosphate in desulfhydrase systems of Proteus morganii.

چکیده
It has been reported (1) that cystine is metabolized by resting cells of Proteus vulgaris and Proteus morganii to pyruvate, ammonia, and hydrogen sulfide, Toluene-killed cells of P. morganii also carried out the reaction and to a lesser degree attacked homocysteine, yielding hydrogen sulfide, ammonia, and some unidentified product. Inasmuch as the use of toluene-killed cells may not obviate entirely considerations of permeability, it was deemed essential to study the reactions with cell-free enzymatic preparations. The present paper describes some of the characteristics of cell-free desulfhydrases in P. morganii.

The L-cysteine desulfhydrase of Escherichia coli.
The coenzyme function of pyridoxal-phosphate in cysteine desulfhydration and in the mechanistically similar serine and threonine dehydration (deamination) processes has been a subject of discussion in a number of recent publications (see, for example, Snell, 1952, and Braunstein and Shemyakin, 1953). A chemical model reaction has been proposed (Metzler and Snell, 1952). Evidence for pyridoxal-p...
متن کاملThe purification and subunit structure of cysteine desulfhydrase from Salmonella typhimurium.
The inducible enzyme, cysteine desulfhydrase, was purified from Salmonella fyphimurium to a state of near homogeneity. This enzyme has a very low tryptophanase activity, lacks cystathionase and tryptophan synthetase activities, and appears to be a specific cysteine desulfhydrase. The purified native enzyme has an s20,W of 10.4 and a molecular weight of 229,000 as determined by equilibrium sedim...
متن کاملEffect of pyridoxal phosphate on atherosclerosis and nephropathy progression in the atherosclerotic rats
Background and Objectives: Diabetes vascular complications are the principal cause of death in the world. Therefore, we investigated the effect of pyridoxal phosphate (PLP) on the formation of atheromatous plaque formation and renal function in the atherosclerotic rats. Materials and Methods: Male Wistar rats were randomly divided into 4 equal groups (ten rats in each group): normal, ath...
متن کاملDeamination of serine. II. D-Serine dehydrase, a vitamin B6 enzyme from Escherichia coli.
Non-enzymatic deamination of serine and cysteine is catalyzed by pyridoxal and certain metal salts at 100” (2). This finding suggested that pyridoxal phosphate might be involved in the enzymatic deamination of these amino acids. Vitamin B, has already been implicated in the desulfhydration of cysteine by rat liver (3) and of cysteine and homocysteine by bacteria (4). Several similarities of cys...
